Effects of light on cell growth and flavonoids biosynthesis in callus cultures of Saussurea medusa Maxim

Abstract
Effects of white light(WL)、blut light(BL)、red light(RL) a nd farred light (FRL) on callus growth, flavonoids biosynthesis and phenylalanine ammonialyase (PAL) activity in the culture of Saussurea medusa Maxim. on solid medium were examined. The results showed that the growth of callus cells was significantly promoted by continuous RL, and moderately by WL and FRL, BL had nearly no effect on the cell growth, compared with that in darkness. However, BL had stronger promotion on PAL activity and flavonoids biosynthesis. The effects of WL on callus growth, PAL activity and flavinoids accumulation were between those of RL and BL. PAL activity and flavonoids accumulation increased as WL fluence rate was raised from 0 to 60#mu#mol m~(-2)s~(-1) while cell growth ch anged little. Experiments with different duration of illumination in a day revealed that PAL activities and flavonoids contents were promoted with the increase of irradiation duration of BL or WL. The experiments with diurnal alteration of WL and BL irradiation indicated that 16h WL/8h BL and 8h WL/16h BL diurnal cycles produced the largest total amount of flavonoids.
